• Transcriptional Regulation of Genomes
Transcription is the first step of gene expression. We are studying at the genome-wide level how the Mediator, a large multi-subunit protein complex conserved through evolution, stimulates transcription by recruiting RNA polymerase II and its general transcription factors on active genes. Our ultimate goal is to understand what are the determinants that govern the pathway of preintiation complexes formation in vivo that lead to transcription activation. We are studying these mechanisms in the yeast Saccharomyces cerevisiae, rodents and Human cells using techniques that range form genetics, molecular biology, biochemistry to functional genomics and high throughput DNA sequencing, and bioinformatics.
